Selective Transformations of Alkylidenecyclobutanes via Ring Opening and Ring Expansion

Selective activation, cleavage, and controllable reassembly of diverse chemical bonds represent a longstanding pursuit in organic synthesis. Strained carbocycles, featuring broad accessibility, thermodynamic stability, versatility, and intrinsic reactivity, have served as indispensable building blocks for the construction of complicated molecular architectures. In the past few decades, selective transformations of alkylidenecyclobutanes and their derivatives through ring-strain-enabled C−C bond activation have been of particular interest owing to the dual reactivity associated with the exocyclic double bond and the cyclobutane motif. This perspective offers a comprehensive summary of the progress pertaining to the ring-opening and ring-expansion reactions of alkylidenecyclobutanes achieved from 2006 to 2026, with a special focus on transition metal catalysis. Selectivity origins, detailed mechanistic dissection, and emerging opportunities are highlighted to spur new reaction design for strained rings.
