Etodolac-loaded mesoporous silica nanoparticles: A novel nanocarrier approach for anti-inflammatory therapy
Introduction Mesoporous Silica, an excipient used enormously for delivering active drug moiety to a specific site. It maintains surface functionality, structure integrity and biocompatibility. Aim and Objective The research was aimed to formulate the mesoporous silica formulation of Etodolac for getting tailored and sustained release of the drug. Material and Method - The formulation was prepared using solvent evaporation method. 2 3 full factorial design was used to carry out optimization and get the most relevant batch. Result and Discussion The finished product was a white to slightly yellowish powder in appearance. The BET Study concluded the filling of the drug into the mesopores of the silica. The optimized batch consisted of % yield 88.82 ± 1.23%, Entrapment Efficiency 74.5 ± 0.31% and Drug loading 60.35 ± 0.68% The particle size, PDI & Zeta potential was obtained 399 nm, 0.519 & −20 mV respectively. These parameters are suitable for oral dosage form. In vivo study was done using carrageenan induced rat paw edema model which signified the inhibition of 86.25 ± 0.18%, proving its effectiveness. Conclusion It was concluded that Etodolac having poor solubility showed increase in drug release pattern when formulated with MSNs. The formulation was further studied for various parameters like drug release, entrapment efficiency, drug content and in-vivo study. Optimized batch was further evaluated for various parameters like DSC study, BET study, Particle size, Zeta potential, SEM analysis and XRD study. The results obtained concluded that the anti - inflammatory effect was increased up to 7 h and it was found effective when compared with marketed drug.
