Doxorubicin and Platinum Nanoparticles Loaded on Hollow Mesoporous Silica for Pancreatic Cancer Treatment

Pancreatic cancer is one of the most invasive malignant tumors with a steadily rising incidence, posing a severe threat to human health. In traditional treatments, surgical resection is the only curative option for early-stage disease, but its insidious early symptoms and lack of specificity often lead to missed optimal treatment opportunities. Chemotherapeutic drugs have poor tumor selectivity, and pancreatic cancer cells show low sensitivity to therapeutics plus high drug resistance, resulting in limited efficacy. Radiation therapy easily damages surrounding normal tissues, highlighting the limitations of traditional modalities. In recent years, nanocarriers have shown great potential in tumor therapy due to their high drug loading capacity, facile functionalization, and favorable targeting. This study constructed a nanotherapeutic platform HMSN@Pt-DOX based on hollow mesoporous silica (HMSN), coloading doxorubicin (DOX), and platinum nanoparticles (Pt) for synergistic chemotherapy and photothermal therapy. Dynamic light scattering revealed a hydrodynamic diameter of 241.14 nm. Cell experiments showed minimal cytotoxicity of the nanocarrier alone (89.5% viability at 200 μg/mL via CCK-8 assay), while combined with photothermal therapy, cell viability dropped to 12.7%. In vivo experiments confirmed that HMSN@Pt-DOX effectively inhibited tumor growth under laser irradiation without obvious systemic toxicity. Leveraging the enhanced permeability and retention (EPR) effect for targeted delivery and near-infrared-triggered local hyperthermia, the platform synergistically enhances cancer cell killing while minimizing side effects. HMSN’s high specific surface area and tunable pores enable efficient drug loading and controlled release. In summary, this nanoplatform significantly improves the therapeutic efficacy against pancreatic cancer and shows potential for inhibiting tumor recurrence.
