Mechanically-Gated Bio-Scaffolds for In-Vivo Hematopoietic Stem Cell Editing: A Dual-Factor "AND-Gate" Architecture for Non-Myeloablative CRISPR Delivery

Current ex-vivo CRISPR therapies for Sickle Cell Disease rely on a highly destructive "system wipe" approach, requiring toxic myeloablative conditioning to clear defective marrow prior to the reinfusion of edited cells. Conversely, systemic in-vivo delivery vectors suffer from rapid hepatic sequestration and off-target payload execution. To bypass these systemic bottlenecks, we propose an implantable, mechanically-gated bio-scaffold that functions as a localized, programmable execution environment a biological "modchip"-for in-vivo hematopoietic stem cell (HSC) editing. This 3D-printed hydrogel matrix utilizes a sustained-release CXCL12 chemokine lure to actively draw circulating, defective HSCs into the enclosed architecture, explicitly bypassing mature, enucleated erythrocytes. To ensure absolute targeting precision, the zwitterionic delivery vehicle is secured by a dual-factor cryptographic hairpin lock. Payload deployment requires simultaneous biochemical verification (CD34+ surface receptor binding) and mechanical activation (25 kPa cellular traction force native to the natural marrow niche). Upon successful integration of the CRISPR construct, the mechanical tether cleanly unzips, allowing the updated HSCs to migrate back into systemic circulation. By establishing a continuous, asynchronous deployment loop, this architecture capitalizes on the extended kinetic survival advantage of wild-type erythrocytes to surpass the 25% systemic stability threshold, effectively reversing the vaso-occlusive phenotype without the need for systemic chemotherapy.
