Solid Supported Synthesis of Different Heterocyclic‐Hydrazone Based Ferrocenyl–cyanovinyl Molecular Hybrids and Investigation of Their Cytotoxicity and Biomolecular Interactions
ABSTRACT Three‐component molecular hybrids involving a heterocyclic moiety, a donor–acceptor redox active cyanovinyl–ferrocenyl unit and a hydrazone linkage, have been synthesized using a novel solid‐state reaction method involving both Schiff‐base and Knoevenagel condensations, stepwise or multi‐component, to explore their biological behavior ranging from cytotoxicity, biomolecular interactions to molecular docking study. Molecular compounds consisting of three different heterocycles, coumarin, pyridyl, and pyrazinyl units, linked to a donor–acceptor redox‐active moiety via hydrazone chain have been investigated to understand their cytotoxicity against THP‐1 cell lines and mode of interactions with BSA and DNA. Molecular docking study showed significant non‐bonding interactions and revealed the role of each component for understanding their potential and future prospect in therapeutic activity. Structural confirmation for the pyridyl‐hydrazone linked cyanovinyl–ferrocenyl molecule has been obtained from single crystal x‐ray diffraction study, while their voltametric evaluation showed relevant redox activity within the molecular system.
