Tri‐ and Tetrasubstituted Allyl Amines: Synthetic Strategies and Developments

Highly substituted allyl amines, particularly tri‐ and tetrasubstituted variants, represent an important yet relatively underexplored class of nitrogen‐containing compounds with broad relevance in pharmaceuticals, natural product synthesis, and functional materials. Their densely substituted olefinic frameworks impart unique steric and electronic features but also create substantial synthetic challenges, including steric congestion and difficulties in controlling regio‐ and E/Z ‐selectivity. While mono‐ and disubstituted allyl amines are readily accessible through classical allylation and hydrofunctionalization methods, efficient and general strategies for highly substituted analogs remain limited. This review highlights recent advances in the synthesis of tri‐ and tetrasubstituted allyl amines, focusing on key developments in transition‐metal catalysis, CH activation, hydrofunctionalization, rearrangement, and cross‐coupling strategies.
