Thermodynamic Anti-Targeting in the Gut-Joint Axis: A Zwitterionic CB1/COX-2 Dual-Inhibitor Scaffold
This paper introduces the MetaboJoint Framework, a novel "Wave-Native" molecular architecture designed to break the pathophysiological feedback loop between systemic obesity and osteoarthritis. The core challenge in treating this comorbidity is the "Asynchronous Compliance Trap": mechanical joint pain prevents the physical mobility required for metabolic weight loss. This research proposes a single-molecule solution—a Bis-Diarylpyrazole Zwitterionic Hybrid—engineered with specific spatiotemporal kinetics. Key Innovations: Thermodynamic Anti-Targeting: Utilizes a permanently charged sulfobetaine zwitterionic linker to create a massive desolvation penalty. This physically forbids Blood-Brain Barrier (BBB) penetration, theoretically eliminating the psychiatric side effects historically associated with CB1 inverse agonists (e.g., Rimonabant). Asynchronous Kinetics: Engineered to provide rapid COX-2 mediated synovial pain relief (Days 1–14) to restore mechanical mobility, followed by sustained, peripherally restricted CB1-mediated metabolic correction (Weeks 2–24). Open-Source Architecture: Provides 2D blueprints, 3D MMFF94-optimized topologies, and the mathematical system dynamics (coupled differential equations) governing the intervention. Goals: This document serves as a formal declaration of Prior Art. The author releases this architecture into the public domain to challenge the open-source scientific community to pursue the synthetic realization and biological validation of peripherally restricted, dual-domain therapeutics.
