Programmable SUST-Based SR-CHA with Ir-Coordination MOFs Enhanced Emitters for Universal Sensitive Evaluation of Various Herpesviruses

The development of a biosensor capable of universal detection for multiple targets has become a critical research direction for overcoming the limitations associated with traditional high-throughput multitarget detection that relies on numerous independent sensing units. A structural unit for simultaneous testing (SUST) capable of target conversion was designed in this study to trigger structural reorganization-catalytic hairpin assembly (SR-CHA), constructing an electrochemiluminescence (ECL) biosensor based on iridium(III) coordination-induced enhanced metal-organic framework emitters for universal detection of herpesvirus on a single system. Impressively, the SR-CHA strategy effectively minimized background noise by optimizing the hairpin structure, leading to high specificity and sensitivity of the biosensor. Additionally, a novel ECL luminophore was synthesized via coordinating an iridium(III) solvent complex with unsaturated nitrogen sites provided by the mixed ligand and metal-organic frameworks. By integrating coordination-induced enhanced strategy and the synergistic enhancement effect of the ligand, the sensitivity was markedly enhanced. Experimental data demonstrated the robust performance of the ECL biosensor in multiplexed detection, which validated the feasibility of the SUST unit trigger SR-CHA strategy for universal detection. Consequently, the sensing strategy proposed in this study further highlighted the application of biosensing in multiplex detection and provided a novel technical pathway toward achieving efficient clinical diagnosis for disease subtyping.
