Frontispiece: Spatial Mapping of Membrane Protein Interactions Using a DNA Origami Rubbing
A strategy named DNA origami rubbing (DOR) was developed for 2D mapping of protein-protein interactions (PPIs) in cell membrane using DNA origami technology and DNA polymerase-mediated strand-elongation, mimicking making stone rubbing with rice paper and graphite. The results showed that DOR can map the fluctuation of 2D distribution of proteins induced by ligands, demonstrating its feasibility for revealing novel PPIs and making it a potential tool for disease screening and predicting medical treatment outcomes.
