Decoding Luciferase Signal Dynamics: A Roadmap to Reliable Luminescent Assays

Luciferases are widely utilized as genetically encodable reporters in diverse biomedical applications including gene expression analysis, biomolecular interaction studies, in vitro diagnostics, and preclinical drug screening. Compared to fluorescent and colorimetric methods, luciferase-based assays offer superior sensitivity, broad dynamic range, and low background interference. However, for decades, the time-dependent nature of luciferase-catalyzed photon emission has been largely overlooked, resulting in compromised quantification and misinterpretation of luciferase activity and cellular reporter assays. In this study, we integrate computational modeling with experimental validation to systematically define the key enzyme kinetic parameters influencing the signal dynamics of commonly used luciferases, particularly those that govern signal intensity and duration, which are critical for reproducible luciferase assays. We demonstrate that ensuring glow-type emission kinetics of luciferase-based luminescence is the key to improving assay quantification, sensitivity, and reproducibility in both in vitro and in cellulo formats. Under optimized assay conditions, we conducted a high-throughput screen of over 8000 bioactive compounds to evaluate nonspecific stabilization and small-molecule interference across benchmarked luciferase reporters. Notably, the recently developed de novo luciferase, LuxSit Pro, exhibited extended signal duration, improved linear dynamic range, and reduced false-positive and false-negative rates compared with luciferases derived from nature. This study provides a comprehensive framework for selecting appropriate luciferase systems, implementing open-access buffer recipes, and applying tailored data analysis strategies to suit diverse experimental objectives. Our roadmap for reliable luminescent assays offers practical guidelines to improve the fidelity, scalability, and versatility of luciferase-based readouts across a wide range of biological and translational applications.
