QbD Driven Targeted Oral Drug Delivery of Famotidine Hydrochloride Loaded Kyron T-114 to Taste Masked Fast Dissolving Tablet Using Ion-Exchange Resin

Introduction: The primary objective of this study was to formulate and evaluate fastdissolving famotidine hydrochloride tablets using an ion-exchange resin to improve patient adherence and enhance dissolution for rapid therapeutic action. Compatibility between the drug and excipients was assessed to ensure formulation stability. Methods: Drug-excipient compatibility was analyzed using FTIR spectroscopy, confirming the absence of chemical interactions. X-Ray Diffraction (XRD) analysis was employed to assess the crystalline nature of the formulations. Pre- and post-compression parameters of the tablets were evaluated in accordance with pharmacopeial standards. The Box-Behnken design was applied for formulation optimization using the desirability approach. Stability studies were conducted under accelerated conditions to determine the robustness of the optimized formulation. Results: The tablets exhibited uniform drug content, low friability, and satisfactory hardness within pharmacopeial limits. The optimized formulation showed rapid disintegration within 16 seconds and achieved 99.5% drug release within 30 minutes. Accelerated stability testing confirmed the maintenance of the drug release profile and physical characteristics for a period of two months. Conclusion: The optimized Famotidine Hydrochloride Fast-Dissolving Tablets demonstrate enhanced dissolution, rapid disintegration, and satisfactory stability. This formulation strategy offers a promising approach to improving therapeutic outcomes and patient compliance in oral drug delivery.
