Oligoarreglo de DNA para detectar los genotipos de virus de papiloma humano 16, 18 y 58

Introduction. Human papillomavirus (HPV) is the etiologic factor for cervical cancer (CC). At present, several HPV tests have been developed using expensive infrastructure and equipment. Objective. To develop and evaluate a single DNA oligoarray for HPV16, -18 and -58 genotypes detection.Material and methods. The design of system detects a small L1/HPV region by recreation of a tandem stable double-strand DNA strategy. This was evaluated for both DNA sources, cervical tissues or cells (fresh) or paraffin-embedded (fragmented), samples.Results. The system, creating a thermodynamically stable molecule at room temperature, successfully detected the HPV16, -18 and/or -58 sequences in both simgle and multiple infection manners after PCR reaction, comparable with DNA sequencing. In addition, it is shown that it is highly specific, sensitive, and able to discriminate between closely related sequences of 20-base length, by using different sources of DNA. The present work provides a reliable and efficient prototype system for HPV detection, showing technically a specific, and sensitive platform. Besides, it could be an important molecular tool for HPV tests in small or medium sized laboratories. Conclusions. This innovative customized system as proof-of-concept research could be used to detect some of the most frequent HPV genotypes, as well as discriminating against different viral loads.
