Epigenetic and transcriptional profiling of secondary hair follicle stem cells during cashmere growth

Cashmere grows from secondary hair follicles (SHFs), which serve as abundant reservoirs of synchronized stem cells responsible for the cyclical regeneration of cashmere. To investigate the epigenetic regulation of this process, we performed ChIP-seq targeting four key histone marks-H3K4me3, H3K27me3, H3K36me3 and H3K79me2 in isolated and cultured secondary hair follicle stem cells (SHFSCs) collected during early anagen and mid-anagen phases. In parallel, RNA-seq was conducted on the same samples to profile transcriptional dynamics. This study provides a genome-wide landscape of histone modifications in SHFSCs and identifies distinct epigenetic signatures associated with the transition from early to mid-anagen. In this Data Descriptor, we detail the experimental design, data generation procedures, quality control assessments, and validation analyses. Together, these datasets offer a comprehensive resource for investigating the epigenetic regulation of SHFSCs and contribute to a deeper understanding of molecular mechanisms underlying cashmere fiber cycling in goats.
