Développement de systèmes d'administration de médicaments auto-microémulsifiants contenant du trans-resvératrol
Trans-resveratrol (t-RSV) is a natural polyphenol with antioxidant and therapeutic potential. However, its poor water solubility significantly limits its oral bioavailability. To improve its solubilisation in aqueous media, self-microemulsifying drug delivery systems (SMEDDS) have been developed. A range of oils, surfactants, and co-solvents were screened to identify the most effective excipients for maximizing t-RSV solubility. Miscibility tests were performed to determine compatible combinations, and various SMEDDS formulations were prepared by varying the oil-to-surfactant-to-co-solvent ratios. The solubility of t-RSV in these formulations was then assessed using UV-Vis spectroscopy, while dispersion tests evaluated microemulsion formation upon water titration. The results obtained demonstrated that an increase in Transcutol® HP content led to a reduction in SMEDDS dispersibility, a phenomenon that was also observed with elevated concentrations of Maisine® CC. The optimal formulations contained at least 10% Transcutol® HP and no more than 10% Maisine® CC. These selected formulations will be loaded with t-RSV and will undergo further dispersibility tests in water to confirm the formation of self-microemulsion and consequently improve solubility and bioavailability of the drug. Preliminary findings suggest that optimized SMEDDS formulations can enhance t-RSV solubility. Further characterization, including particle size analysis and stability studies, is currently underway to validate their suitability as an oral delivery system.
