Determination of extraction conditions for triterpenic compounds of Prunus domestica l. fruit samples

Introduction. Extraction is a key step in isolating bioactive compounds from plants. Optimizing factors like solvent ratio, time, and composition is essential to maximize triterpenic compound yield [2]. Materials and methods. 1 g of lyophilized Prunus domestica L. fruits powder was placed in dark glass bottles, covered with 10 ml 100% (v/v) of acetone and placed in an ultrasonic bath. The ultrasonic bath operates at a frequency of 80 kHz. The extracts were filtered through a vacuum pump and 0.22 μm membrane filters before HPLC analysis [1]. The results were analyzed using 'MS Excel 2023' ('Microsoft', Redmond, Washington, USA) and 'SPSS Statistics 21' ('IBM', Chicago, Illinois, USA). Results ant their discussion. In the first stage, the impact of different extraction solvents on the recovery of triterpenic compounds from P. domestica fruits was assessed. The selected extraction solvents included 96% ethanol (v/v), 100% methanol and 100% acetone (v/v). Acetone produced the highest extraction yield (81.72±3.70 µg/g), and therefore was selected for further experiments. In the next stage, the impact of extraction time (ranging from 10 to 60 minutes) was assessed, with the best yield (84.40±1.56 µg/g) achieved at 50 minutes. Finally, the effect of ultrasonic power on extraction efficiency was analyzed, with the best result (96.26±4.93 µg/g) obtained using 791 W ultrasonic power. Summarising the results, these extraction conditions could be used for the further researches of triterpenic compounds of P. domestica fruits.
