Dual-Responsive Hyaluronic Acid-Modified Zeolitic Imidazolate Framework-8 Nanoplatform for Targeted Antibacterial Therapy and Wound Healing
In daily life, the skin is susceptible to damage and secondary bacterial infections. However, traditional treatments have difficulty simultaneously inhibiting bacteria and promoting wound healing. To address this challenge, we developed a multifunctional nanoplatform, ZIF-8@NFX@HA (ZNH). The platform utilizes pH-responsive zeolitic imidazolate framework-8 (ZIF-8) loaded with norfloxacin (NFX) and modifies the outer layer with hyaluronic acid (HA) through coordination and electrostatic interactions. ZNH is dual-responsive, capable of targeting drug release according to the pH value and hyaluronidase activity of the infected microenvironment, ensuring precise antibiotic delivery. In addition, ZNH could accelerate wound healing by promoting the production of collagen and neovascularization. Experimental findings in vitro and in vivo demonstrated that ZNH possessed potent bactericidal efficacy against both Gram-negative and Gram-positive bacteria, as well as good biocompatibility. ZNH also exhibited excellent therapeutic efficacy in a murine skin infection model. This work provides a promising strategy for managing wound infections.
