A new hydroxybutanamide derivative from <i>Saussurea involucrata</i> with anti-PD activity on H 2 O 2 insult sH-SY5Y cells

A new hydroxybutanamide derivative, involucramide (<b>1</b>), was obtained from the traditional Chinese medicinal herb <i>Saussurea involucrata.</i> Five other isolates were characterised as betulinic acid (<b>2</b>), costunolide (<b>3</b>), dihydrocostunolide (<b>4</b>), isodihydrocostunolide (<b>5</b>), and dehydrocostus lactone (<b>6</b>). The structure of <b>1</b> was established through extensive data analyses encompassing its NMR, IR, UV, and HRESIMS data. All isolates were evaluated for the anti-PD activity on H2O2-insulted SH-SY5Y cells. Amongst them, compound <b>1</b> exhibited the potential activity with the EC50 value of 6.84 <i>µ</i>M. Furthermore, at the concentration of 20 <i>µ</i>M, <b>1</b> attenuated the intracellular ROS accumulation, elevated the glutathione (GSH) levels, and reversed the downregulated mRNA expression of HO-1 and NQO1 in H2O2-injured SH-SY5Y cells.
