Efficient Synthesis of Fused 1,2,4-Triazolothiadiazole Derivatives: A Green and High-Yield Approach and Biological Evaluations

An optimized synthetic approach has been developed for fused 1,2,4-triazolothiadiazole derivatives under environmentally benign conditions using a green solvent and microwave irradiation technique. The proposed methodology enhances reaction efficiency, reduces byproduct formation, and improves overall yield compared to conventional synthesis. Structural characterization of the synthesized compounds was performed through NMR, FTIR, and mass spectrometry. Reaction optimization studies have shown that catalyst selection, reaction temperature, and solvent choice significantly impact product yield and purity. Comparative analysis with traditional methods highlights the advantages of the proposed strategy in terms of sustainability, atom economy, and scalability. Biological evaluation of the synthesized fused 1,2,4-triazolothiadiazole derivatives showed their potent antimicrobial and anticancer activities, with selected derivatives exhibiting superior efficacy compared to standard drugs. The structure–activity relationship (SAR) analysis revealed that specific functional groups significantly influence bioactivity, providing insights into future molecular modifications.
