Variations characteristics of triglyceride during various stages of ovarian development in commercial production of Penaeus monodon broodstock

This study investigated the role of triglyceride (TG) metabolism in ovarian development of Penaeus monodon broodstock. Female shrimp were fed with the model of “food organisms+commercial feed” commonly used in actual production. Serum, hepatopancreas, and ovary samples were taken at the pre-induction stage (prior to unilateral eyestalk ablation), as well as stages I, III, and V of ovarian development. The results showed that from pre-induction stage to stage I, TG content significantly increased in serum and ovary but remained stable in hepatopancreas. The hepatopancreas showed increased SFA and PUFA but decreased MUFA (all belong to free fatty acids, FFAs), while the ovary exhibited rises in these FFAs. Ovarian TG synthesis genes ( fas , srebp1 , gpat3 , mogat2 , dgat1 , lpin3 , acc , scd5 and gpd1 ) were up-regulated, while hepatopancreatic expression of gpd1 , lpin3 and dgat1 declined, and mogat2 , srebp1 , acc and scd5 increased. Ovarian TG catabolism genes ( hsl , mgl , daglβ and dgk ) were elevated, and hepatopancreatic hsl and dgk increased, but mgl decreased. TG transport genes ( mttp and ldlr1 ) were up-regulated in ovary, and hepatopancreatic mttp enhanced. From stage I to V, TG contents in serum and hepatopancreas, the above-mentioned FFAs in hepatopancreas exhibited a tendency of first reducing and then increasing, whereas the above indicators in the ovary showed a trend of first growing and then decreasing. Furthermore, genes expression related to TG synthesis, catabolism, and transport exhibited stage- and tissue-specific patterns, highlighting distinct metabolic partitioning between tissues. In conclusion, adequate supplementation of TG is essential for supporting ovarian development in shrimp broodstock. • This study elucidates the dynamic changes in TG metabolism during shrimp ovarian development. • A large amount of TG is transported from the hepatopancreas to the ovary during stages I to V. • The demand for free fatty acids in shrimp ovaries peaks during stage III.

Abstract
