18 01 Base Architecture Triple-Negative Breast Carcinoma TNBC
Triple-negative breast cancer (TNBC) accounts for 1520% of breast malignancies yet contributes disproportionately to mortality due to aggressive biology and limited targeted options . The defining characteristic—absence of estrogen receptor (ER), progesteronereceptor (PR), and HER2 amplification—renders conventional endocrine and HER2-targeted therapies ineffective.Current standard-of-care relies on cytotoxic chemotherapy with 5-year survival rates of 40-50% in advanced disease .Recent advances in tumor ecology reveal TNBCas a spatially heterogeneous ecosystem with distinctmetabolic niches . Hypoxic regions drive glycolyticreprogramming and lactate accumulation, while perivascular zones maintain oxidative metabolism . Cancerstem cells residing in protective niches exhibit profoundtherapy resistance through metabolic plasticity, redoxbuffering, and quiescence. This model tries to make a snaphot of the carcinomas base architecture and dynamic .
