Effect of Platelet Rich Plasma on Male Reproductive Organs Toxicity Induced by Sodium Valproate in Rats

Background: Sodium valproate (SVP) is an anticonvulsant and mood-stabilizing medication widely used in managing epilepsy, bipolar disorder, and migraine prophylaxis. Despite its effectiveness; it has potential adverse effects on testicular function. Platelet-rich plasma (PRP) is a five-fold higher platelet content concentrated blood product. PRP became important in regenerative medicine. Its rich content of growth factors enhance cellular growth, differentiation and proliferation. Materials and Methods: The study evaluated PRP’s therapeutic effects on SVP-induced testicular toxicity. PRP was prepared from healthy rat’s blood and biochemically analyzed. Seventy-two adult male rats were segregated into two groups: healthy and toxicity-induced. Rats were administered SVP (400 mg/kg) via intragastric tube for two weeks to induce testicular toxicity. After confirming toxicity, main groups were divided into six subgroups. Group 1: healthy rats were given distilled water (DW) and injected by saline solution, group 2: rats were given SVP (400 mg/kg) day after day and injected by saline solution , group 3 and 4: healthy rats were given DW and injected by 0.5 ml/kg and 1ml/kg PRP twice weekly , respectively , group 5 and 6: rats were given SVP (400 mg/kg) day after day and injected by 0.5 ml/kg and 1 ml/kg PRP twice weekly , respectively . Results: PRP significantly counteracted infertility by enhancing sperm parameters, fertility hormones, antioxidant status, testicular integrity and improving testicular tissue histological architecture while decreasing inflammation and oxidative stress. Conclusion: Results concluded that administration of PRP attenuated the changes induced by SVP consumption that can lead to infertility.
