Comparative Formulation and Solubility Assessment of Beta Cyclodextrin-Based Nanosponges Containing DHA and Aceclofenac

AbstractDHA and Aceclofenac show poor water solubility, limited stability, and weak oral absorption. These traits reduce their therapeutic value and create barriers for formulation work. Betacyclodextrin based nanosponges offer a structured system that improves solubility, stability, andrelease behavior. This chapter gives a detailed comparison of DHA loaded and Aceclofenacloaded nanosponges. It covers formulation design, preparation methods, characterization steps, solubility assessment, release behavior, and stability outcomes. The goal is to explain hownanosponge networks modify drug performance and to highlight key differences between the twomolecules.DHA is a long chain fatty acid. It oxidizes fast and stays poorly soluble in water.Aceclofenac is a weakly acidic NSAID with limited dissolution at physiological pH. Both drugsneed better solubility and controlled release to reach effective plasma levels. Beta cyclodextrinsupports these improvements because it forms host–guest inclusion complexes through itshydrophobic cavity. Cross linking converts cyclodextrin molecules into a three dimensionalporous nanosponge network. This network traps drug molecules and shields them fromdegradation while improving wettability and dispersibility.Formulation work used solvent basedand ultrasound assisted methods. Solvent based preparation produced uniform nanosponges withpredictable morphology. Ultrasound assisted preparation reduced processing time and improvedparticle size control. Both methods allowed stable loading of DHA and Aceclofenac.Optimization steps focused on the ratio of beta cyclodextrin to cross linker, drug to polymerratio, pH conditions, and stirring duration. These factors influenced particle size, entrapmentefficiency, and drug release behavior.Characterization confirmed nanosponge formation. Particlesize analysis showed values in the nanometer range with narrow PDI. Zeta potential indicatedstable colloidal behavior. SEM and TEM produced clear images of porous spherical structures.FTIR confirmed drug incorporation through shifts in key functional group peaks. XRDdemonstrated reduced crystallinity of the drugs after loading, which contributed to enhancedsolubility. DSC and TGA provided thermal profiles that showed improved stability of DHA andAceclofenac within the nanosponge matrix.High Technology Letters Drug loading and entrapment efficiency differed between the two drugs. DHA showed strongerinteraction with beta cyclodextrin, driven by its hydrophobic nature and flexible chain structure. This produced higher loading and higher entrapment efficiency. Aceclofenac demonstratedmoderate interaction levels. It formed stable complexes but showed lower affinity than DHA.These differences influenced solubility enhancement and release profiles.Solubility studiesshowed significant gains for both drugs. DHA exhibited a marked increase in aqueous solubility, with stable dispersion in water for extended periods. Aceclofenac showed moderateimprovement, especially in buffer systems near neutral pH. Higher nanosponge concentrationfurther improved solubility due to increased availability of inclusion sites. The solubility shiftsupported better dissolution and uniform release behavior during in vitro tests. In vitro releasestudies used standard dissolution conditions. DHA nanosponges showed sustained release withreduced burst effect. The release pattern depended on cross linking density and preparationmethod. Aceclofenac nanosponges released at a slightly faster rate due to weaker bindinginteractions. Both drugs displayed higher release amounts than their pure forms. Releasemodeling supported diffusion controlled mechanisms with modified matrix erosion.Stabilitystudies assessed physical and chemical stability under stress and accelerated storage conditions.DHA showed improved resistance to oxidation due to protection from the nanosponge network.Aceclofenac maintained structural integrity without major degradation peaks. Particle size andzeta potential remained stable throughout the study period. These findings confirm thatnanosponge encapsulation increases shelf life and prevents rapid degradation.Comparativeanalysis highlighted the influence of drug structure on nanosponge performance. DHA, with highhydrophobicity, interacted strongly with the cyclodextrin cavity and the polymer network. Thisproduced higher loading, greater solubility enhancement, and stronger stabilization. Aceclofenacinteracted less strongly but still gained improved dissolution and controlled release. Thesedifferences show the ability of nanosponges to adapt to diverse molecular structures.
