In vitro Pseudomonas aeruginosa and its association with the stimulation of pro-inflammatory gene expression in mouse immune cells
The aims of this study determined to investigate the inflammatory immune response induced by Pseudomonas aeruginosa in immune cells in vitro .In this study , an immune cells were isolated from secondary immune organs of both spleen and lymph nodes . Cells were divided into two groups in each organ include control inactivated group and activated group which include Pseudomonas aeruginosa that was cultured with immune cells. After 24 hrs. of incubation in CO2 incubator cells were collected then molecular tests were done to investigate immune response by measuring the fold change of targeted inflammatory immune genes include CD3, IL-2, TNF α and NF kappa B . The results of our study in vitro indicating that Pseudomonas aeruginosa was able to induce upregulation in the fold change of all markers in both spleen and lymph node with arrange between 3 folds to more than 47 folds compared to one fold only in the control group, our results suggested that Pseudomonas aeruginosa has the ability to induce the proinflammatory genes in the immune system though activation of the immune molecular pathway signaling in vitro.
