Epigenetically Supported Human Longevity: Coordination of Natural Signaling Pathways
This preprint proposes and elaborates the concept of "epigenetic synchronization" — a systems-level framework for understanding how modifiable lifestyle and environmental factors may influence DNA methylation–based biomarkers of biological aging. The preprint synthesizes evidence on core longevity pathways (AMPK/SIRT1, mTOR, NRF2, FOXO) and key contextual modulators, including circadian stability, inflammation, oxidative stress, and the exposome. It critically discusses the interpretational limitations of epigenetic clocks and outlines testable hypotheses for future research. This synthesis provides a scientific framework for research into interventions that may support healthy aging and the potential for rejuvenation of epigenetic biomarkers. Keywords: epigenetic clocks, DNA methylation, AMPK, SIRT1, mTOR, NRF2, FOXO3, circadian rhythms, exposome, inflammation, epigenetic synchronization, biological age, longevity, wellness.
