Chromosome-level genome assembly and annotation of the endangered plant Primula kwangtungensis (Primulaceae)

Primula kwangtungensis, a perennial herbaceous plant endemic to the Nanling Mountains in southern China, is rare and sparsely distributed, making it a conservation priority. This study reports a high-quality chromosome-level genome assembly of approximately 1.29 Gb, achieved through BGI-DIPSEQ, Nanopore, and Hi-C sequencing. Hi-C data anchored 88.68% of contigs to nine chromosomes, with contig N50 of 5.87 Mb and scaffold N50 of 144.82 Mb. The assembly achieved a BUSCO completeness score of 95.8%, indicating high accuracy. A total of 31,717 protein-coding genes were identified, with 97.46% functionally annotated, and repetitive sequences accounted for 63.25% of the genome. This assembly provides crucial data for understanding species diversity mechanisms in the Nanling region and the comparative genomics studies among Primulaceae. It offers essential genetic resources for biodiversity research and aids in developing conservation strategies for rare and endangered plant species.
