Catalytic Asymmetric Functionalization of 4‐Methylcoumarins: A Vinylogous Nucleophile Perspective

The broad utility of coumarin scaffolds has long intrigued organic chemists, driving extensive research into their synthesis and functionalization. Over the decades, functionalized coumarins have emerged as privileged frameworks in natural products, biologically and pharmaceutically relevant molecules, as well as in dyes and advanced materials. While numerous strategies have been developed for C-H functionalization at the C-3, C-4, and C-7 positions through organo-, metal-, and photocatalytic methods, the pursuit of remote C-H functionalization in coumarin analogues, particularly as an alternative mode of activation that harnesses their vinylogous nucleophilicity remains both compelling and challenging. This perspective provides a comprehensive overview of recent catalytic advances in the asymmetric remote C-H functionalization of 4-alkylcoumarins as pro-vinylogous nucleophiles. It also highlights future directions for the development of efficient catalytic systems to unlock novel transformations, underscoring their untapped potential in medicinal chemistry.
