3D Pharmacophore Mapping: A Robust Framework for Virtual Screening in Modern Drug Discovery
Abstract: Spatial arrangements and chemical features of molecules play a pivotal role in drug discovery. These features directly influence the interaction between a drug candidate and its biological target. Pharmacophore mapping is a powerful technique that allows researchers to identify and characterize these critical features, facilitating the design and optimization of effective therapeutics. This review provides a comprehensive overview of 3D pharmacophore mapping features, methodologies, and the validation of pharmacophore hypotheses based on receptor-ligand interactions. Additionally, it describes the applications of 3D pharmacophore mapping in various therapeutic areas, highlighting case studies that demonstrate its effectiveness in identifying new drug candidates. By streamlining the virtual screening process, this framework not only accelerates drug development timelines but also minimizes costs associated with experimental validation. Our findings underscore the significance of 3D pharmacophore mapping as a transformative tool in modern drug discovery, paving the way for the development of innovative therapeutics tailored to specific biological targets.
