Isolation of Flavonoid Bioactive Scutellarin and Hispidulin from Scutellaria barbata D. Don Herb

Introduction: Scutellaria barbata D. Don, a traditional medicinal plant, has attracted growing scientific interest due to its diverse pharmacological activities. Methods: The present study focuses on the isolation and structural characterization of two bioactive flavonoids, hispidulin and scutellarin, from the hydroethanolic extract of S. barbata. Extraction was performed by cold maceration using 70% hydroethanol, followed by sequential fractionation with solvents of increasing polarity (n-hexane, dichloromethane, ethyl acetate, and nbutanol). Results: The ethyl acetate fraction was selected based on preliminary phytochemical screening and TLC profiling, leading to the successful isolation of hispidulin (5% yield) and scutellarin (11% yield). High-purity compounds were obtained through column chromatography, and their identity was confirmed using complementary analytical techniques, including TLC, UV spectroscopy, LC-MS, and NMR. Discussion: The results provide reproducible methodological details and confirm the purity and structural features of the isolated compounds. Conclusion: Overall, this study underscores the pharmacological potential of hispidulin and scutellarin and provides a reliable framework for future therapeutic exploration of S. barbata-derived bioactives.
