67eP A two-gene signature based on HOXA5 and CEACAM16 predicts survival in IDH-Mutant glioma

Background: In the battle against the world's deadliest cancer, lung cancer, immunooncology (IO) treatments have increased the survival of many patients. However, treatment resistance remains the most common outcome. This study aims to address this issue by examining the peripheral immune response in Non-Small Cell Lung Cancer (NSCLC) patients undergoing IO treatment. Methods:We are currently collecting PBMCs, blood plasma, and serum from these patients.Using this material, we are conducting multilevel multi-omics analyses at the single-cell level to identify diagnostic and prognostic blood-based biomarkers. Our goal is to discover biomarkers that can predict treatment outcomes, guide the personalization of NSCLC treatments, and reveal new therapeutic targets.Additionally, we are investigating correlations between PBMC composition and the diagnostic details of the tumor microenvironment. Results:Preliminary results have identified several potential blood-based biomarkers that correlate with tumor features such as PD-L1 expression, metastatic status, and smoking history. In parallel with ongoing cohort collection, we are analyzing singlecell transcriptomics and spectral flow cytometry data from PBMCs of patients receiving IO treatment. Conclusions: These emerging correlations may help reduce the reliance on invasive lung biopsies by providing minimally invasive diagnostic alternatives.
