322TiP ENCEFALO (GEM2301) clinical trial: Encorafenib and binimetinib followed by cemiplimab and fianlimab in BRAF-mutant melanoma and symptomatic brain metastases

Patients (pts) with BRAF-mutant melanoma symptomatic brain metastases (MBM) have a high unmet medical need. Targeted therapy (TT) has shown high intracranial response rates (RR) in this population independent of the symptomatic status. Encorafenib (E) and binimetinib (B) showed an ORR over 60% in pts with symptomatic brain metastasis. However, the duration of responses with TT remains short-termed in most cases. Conversely, immunotherapy (IT) shows low RR but is capable of achieving long-term disease control, mostly in asymptomatic pts.
