Directional Pressure Failure: A Unifying Orientation for Insulin Signaling, Buffering Failure, and GLP-1 Response, Lantern of Sulfur, Concept B0, v12
This Concept B0 note provides a unifying orientation for Directional Pressure Failure (DPF). It clarifies how insulin signaling, cellular hydration, electrolyte buffering, bile-mediated clearance, and GLP-1 receptor agonist response nest within a single directional law. This document does not propose new mechanisms or expand the core DPF claim. Its purpose is to formalize the interpretive hierarchy: inward pressure vectors (e.g., insulin-driven substrate uptake and GLP-1–amplified signaling) become destabilizing when downward buffering and clearance are unavailable, as in hyperchloremia with non–anion gap metabolic acidosis (NAGMA). By naming this hierarchy explicitly, B0 is designed to prevent misclassification of DPF phenomena as isolated metabolic, dietary, or pharmacologic effects. Subsequent Concept B extensions apply this framework to specific physiologic terrains, including insulin signaling and cellular hydration in hyperchloremia with NAGMA (Concept B5). In DPF terrain, signal amplification becomes load amplification whenever downward exits (buffering + clearance) are constrained. Navigation: Master Index and Conceptual Map.
