A3 coupling of 1,2,3-Triazoles: Microwave-enhanced catalysis with [pyridine–SO3H]CuCl3

This study introduces a microwave-enhanced catalytic system for A 3 coupling to synthesize 1,2,3-triazoles using the novel [Pyridine–SO₃H]CuCl₃ catalyst, a copper-based complex featuring a pyridine-sulfonic acid ligand chosen for its unique ability to promote this reaction. Responding to the push for sustainable and efficient heterocyclic synthesis, the work targets the pharmaceutically important 1,2,3-triazole scaffold by integrating microwave irradiation with a purposefully designed catalyst to achieve rapid, high-yielding coupling under mild conditions. The catalyst was synthesized and thoroughly characterized by FT-IR, TGA, XRD, and EDX, confirming its structural features. Under optimized microwave conditions, the system enables the three-component coupling of aldehydes, alkynes, and amines to deliver 1,2,3-triazoles with exceptional efficiency, demonstrated across eighteen substrates with yields in the 90–95 % range and broad functional-group tolerance. Notably, reaction times are markedly shorter than those obtained under conventional heating, highlighting the kinetic advantages of microwave irradiation. The methodology offers rapid reaction times, uniformly high yields, mild conditions, straightforward workups, excellent functional-group tolerance, and catalyst recyclability for at least five cycles, underscoring its practicality for repeated use. Mechanistic insights indicate that the catalyst simultaneously activates the alkyne and aldehyde, enabling the formation of the copper acetylide intermediate; the synergy of microwave acceleration with this tailored catalyst yields substantial time savings while maintaining product quality. Collectively, the work represents a meaningful advance in 1,2,3-triazole synthesis, offering a practical, efficient route with broad substrate compatibility and significant potential for applications in medicinal chemistry and materials science, where rapid access to these heterocycles is advantageous. Abstract
