Effets du déficit en NRP1 sur la production de rénine et la fibrose rénale médiée par les fibroblastes
NRP1 is a transmembrane co-receptor that was first found in the neural system. It can react with different growth factors, such as class 3 Semaphorins, VEGF, and TGFβ. NRP1 can play a role in multiple biological and pathological processes, including neuronal development, angiogenesis, and tumor progression. In the kidney, Nrp1 can be expressed in many kinds of cells. But the role of Nrp1 in renal physiological and pathophysiological is remains unclear. To figure out what is the role of Nrp1 in the kidney, we constructed the Nrp1-KO mice as the experimental group, compared the renal structure and function under physical conditions and sodium homeostasis unbalanced conditions, in addition, we compared the severity of renal fibrosis with the renal fibrosis mouse model and what is the reason to make the difference. We found that under basal conditions, the renal structure and function are not influenced when lacking Nrp1. When facing the sodium homeostasis imbalance, the ability to adapt to the imbalance is worse, and the renin in JG cells is less detected. When under fibrotic conditions, Nrp1-KO has more severe renal fibrosis, the reason is probably that Nrp1 influences the tissue repair of fibroblasts when in the acute injury phase. What's more, Nrp1-KO mice even have more severe cardiac fibrosis, which means the protective function of Nrp1 can apply to other organs.
