Novel Renal-Protective Adjuvants in Immunosuppression: A Systematic Literature Review

Immunosuppressive therapies, although essential for treating autoimmune disorders and averting organ rejection, frequently cause substantial kidney damage, which underscores the need to investigate adjuvants that reduce such harmful effects without diminishing therapeutic effectiveness. This systematic review evaluates the existing evidence on innovative renal-protective adjuvants included in immunosuppressive protocols, with particular attention to their mechanisms, clinical results, and capacity to safeguard kidney function. A thorough examination of peer-reviewed research was performed to pinpoint adjuvants working in concert with immunosuppressive drugs, especially in scenarios including transplantation, autoimmune diseases, and cancer immunotherapy, where safeguarding kidney function is critical. The results indicate that specific adjuvants, such as immunomodulatory molecules and targeted drug delivery systems, show potential nephroprotective effects by reducing oxidative stress, inflammation, and drug-induced nephrotoxicity. Notably, vaccines and adjuvants developed to regulate immune responses display dual advantages in improving therapeutic outcomes while diminishing renal injury, although their clinical feasibility is still being studied. The review also identifies shortcomings in translational research and stresses the necessity of rigorous clinical trials to substantiate preclinical findings. Collectively, these insights underscore the potential of renal-protective adjuvants to refine immunosuppressive strategies, thereby improving patient outcomes in high-risk populations. Future studies ought to focus on mechanistic investigations and extended safety evaluations to support their inclusion in standard treatment guidelines.
