The Palade Conjecture The supremacy of MaRS in clinical detection of Familial Hypercholesterolemia of young adults between 30 and 45 years and the Triumph of Palade FH detection.pdf
This paper introduces the Palade Conjecture, a theorem linking clinical and genetic early detection of familial hypercholesterolemia (FH) in young adults (30–45 years). Building on the Marciu Risk Score (MaRS) — a non-laboratory predictive framework capable of identifying 50–70% of FH cases — the study defines Palade FH Detection, a novel genetic screening concept honoring George Emil Palade, Nobel laureate in Physiology or Medicine. Through validated datasets (JACC 2019, EAS 2022), the work proposes that early genetic screening for LDLR, APOB, and PCSK9 mutations can reduce preventable cardiovascular mortality by establishing a direct correspondence between simulated prevalence and real-world survival outcomes. The Palade FH Cohort Iterator formalizes epidemiological bijectivity — transforming the implicit extrapolation from cohort data to population-level prevalence into a verifiable and reproducible model. Together, MaRS (an Eulerian weak hypothesis) and Palade FH Detection (a Goldbachian strong hypothesis) form a dual framework bridging predictive modeling with clinical genetics, redefining FH prevention strategies worldwide.
