From Data to Decision A Case-Based Statistical Analysis of Pharmaceutical Stability per ICH Q1E

Abstract The stability of pharmaceutical drug products is a cornerstone of quality assurance, safety, and therapeutic efficacy. Regulatory bodies such as the FDA, WHO, ICH, and EMEA mandate that every marketed drug product must display a scientifically justified expiration date on its immediate container label. This requirement underscores the critical role of stability studies in determining shelf-life, especially for newly developed and generic formulations. Statistical evaluation, as outlined in the ICH Q1E guideline, provides a robust framework for interpreting stability data and estimating product longevity under various storage conditions. This paper presents a comprehensive case study that integrates both theoretical foundations and practical applications of statistical methodologies in stability analysis. Key statistical tools—including regression analysis, analysis of variance (ANOVA), residual diagnostics, p-values, and coefficient of determination (R²)—are employed to assess assay values and degradation trends over time. The study also explores the use of shelf-life plots and multiple response prediction models to enhance the precision of expiration dating. By applying these techniques within the ICH Q1E framework, the paper demonstrates how statistical thinking ensures reliable, reproducible, and regulatory-compliant shelf-life estimations. The findings highlight the importance of data integrity, model selection, and diagnostic validation in pharmaceutical stability studies. Ultimately, this research reinforces the indispensable role of statistics in bridging laboratory data with regulatory expectations, contributing to the safe and effective delivery of drug products to patients worldwide.
