Risk–Benefit Trade-offs of GLP-1RAs and SGLT-2is in Type 2 Diabetes Mellitus (T2D): Systematic Review, Meta-Analysis, and Net Benefit Modelling

This dataset supports a systematic review and meta-analysis evaluating the efficacy and safety of glucagon-like peptide-1 receptor agonists (GLP-1RAs) and sodium-glucose cotransporter-2 inhibitors (SGLT-2is) in adults with type 2 diabetes mellitus (T2DM). The study synthesizes data from 17 cardiovascular outcome trials (CVOTs) involving 132,038 participants (73,263 GLP-1RA, 58,775 SGLT-2i), sourced from PubMed and Cochrane CENTRAL up to September 14, 2025. It includes search strategies, screening decisions, extracted data, and statistical outputs for major adverse cardiovascular events (MACE), hospitalization for heart failure (hHF), renal composite outcomes, and safety outcomes (e.g., retinopathy, genitourinary infections, diabetic ketoacidosis, amputations). Novel features include cross-class comparisons, net benefit analyses (absolute risk reductions minus absolute excess risks), and graphical syntheses (forest, tornado, scatter plots, risk curves). The dataset contains: (1) raw search outputs from PubMed (16,651 records) and Cochrane CENTRAL (210 records); (2) Rayyan screening logs (1,098 titles/abstracts, kappa=0.89); (3) Excel files with extracted trial data (study characteristics, participant demographics, HRs/RRs, event rates); (4) R scripts for random-effects meta-analyses; and (5) supplementary tables and figures. Data were collected by Dr. Samit Ghosal (conceptualization, searches, analyses) and Anuradha Ghosh (search verification, manuscript assistance). The study is registered on PROSPERO (CRD42024567890). This dataset enables replication and further exploration of cardiorenal outcomes in T2DM management
