Evaluation of the PAT Gene as a Selectable Marker for Genetic Transformation in Scenedesmus obliquus
The utility of the phosphinothricin acetyltransferase (PAT) gene as a selectable marker was evaluated for its effectiveness in genetic transformation and stable production of the microalga Scenedesmus obliquus. The PAT gene, conferring resistance to herbicides like glufosinate ammonium, offers a robust selection system that facilitates the isolation of transformed algal cells. In this study, S. obliquus was transformed via an optimized method using the PAT gene, and its functionality was assessed under varying concentrations of selective agents. The results demonstrated high transformation efficiency, stable gene integration, and consistent expression of the selectable marker. Furthermore, the transformed cells maintained normal growth and photosynthetic activity, indicating minimal metabolic burden from PAT gene expression. These findings highlight the potential of the PAT gene as a reliable tool for genetic engineering in S. obliquus, paving the way for advancements in biofuel production, carbon capture, and other biotechnological applications.
