Real-Time, Closed-Loop RNA Virus Detection and Therapeutic Delivery via CRISPR-Cas13d-Mediated Programmable Nanobody-Conjugates
**Abstract:** We propose a novel platform for real-time detection and targeted therapeutic delivery against RNA viruses leveraging the programmable specificity of CRISPR-Cas13d and the precision of engineered nanobody-conjugates. This system, termed "ViruLock," dynamically monitors viral RNA presence, initiating therapeutic intervention within minutes of detection. The innovation lies in integrating a highly sensitive Cas13d-mediated RNA detection reporter with a biocompatible, modular nanobody-drug delivery scaffold, enabling closed-loop viral control. The platform offers a significant advancement over existing diagnostics and treatments, potentially allowing for rapid and personalized intervention against emerging viral threats, increasing treatment efficacy and minimizing off-target effects.
