PCC1 Senolytic Complex Compound's Preliminary Study on Regulating Cellular Senescence
Cellular senescence significantly impacts organismal aging and related diseases through the accumulation of senescent cells and their pro-inflammatory senescence-associated secretory phenotype (SASP). Procyanidin C1 (PCC1) is a natural trimeric compound extracted from grape seeds, which has demonstrated selective clearance of senescent cells and anti-inflammatory effects in animal models; however, its oral bioavailability is limited, hindering clinical translation applications. Currently, there are no reports on the effects of PCC1 on human aging. Here, we present the first retrospective study investigating the impact of PCC1 senolytic complex (PCC1-SC) products in elderly individuals. This phytosomal formulation enhances gastrointestinal absorption by 2- to 3-fold, addressing the bioavailability barrier. Following a three-day oral nutritional intervention, 17 participants aged 50 to 86 exhibited a marked reduction in senescence biomarkers, including p21^CIP1 and SA-β-Gal, in peripheral blood mononuclear cells, with no observed differences between genders. Concurrently, plasma levels of several key SASP factors—IL-6, IL-8, VEGF, and TGF-β1—were significantly reduced. Importantly, no adverse events were reported, underscoring the favorable safety profile of this intervention. This investigation for the first time demonstrates that PCC1 has the activity of selectively clearing senescent cells and systemically inhibiting SASP in the elderly population, providing support for its further development as a next-generation anti-aging therapy.
