Biotechnology: embryoculture of steppe cherry (Prunus fruticose Pall.)
The results of a study on microclonal reproduction of steppe cherries are presented. The literature data on methods of sterilization and scarification of steppe cherry seeds are reviewed. The purpose of the research is to analyze proven approaches and select optimal conditions for obtaining a culture of steppe cherry varieties for further work on creating a living collection in vitro and improving the methods of embryoculture and sterilization. Two varieties of steppe cherries were studied: Shakirovskaya and Zamenka, as well as plants numbered 5-88 and 2-230. The planting material of the steppe cherry varieties was provided by the Sverdlovsk Horticulture Breeding Station, a structural unit of the Federal State Budgetary Scientific Institution of the Ural Branch of the Russian Academy of Sciences. In total, 86 explants of plants of the Shakirovskaya variety were planted, 209 of the Zamenka variety, 168 of the 2-230 sample and 117 of the 5-88 sample. The technique was based on classical methods of working with cultures of isolated plant tissues and organs: preparation of nutrient media, sterilization of instruments and equipment, and work in aseptic conditions. The research was conducted in 2025. at the Department of Botany and Plant Genetics of Perm State National Research University. As a result of the conducted research, the modes of sterilization of explants and variants of Murashige and Skuga nutrient media for the introduction of steppe cherries into culture in vitro were selected. The percentage of surviving samples varied depending on the genotype. The results of the work will be used in the formation of a collection of cherry varieties in vitro.
