Automated Variant Annotation and Prioritization for Ultra-Rare Mendelian Disease Diagnostics via Multi-Modal Knowledge Fusion
**Abstract:** Accurate and efficient variant annotation and prioritization are crucial for diagnosing ultra-rare Mendelian diseases. Current approaches struggle with the complexity of these diseases, often requiring substantial manual curation and generating numerous candidate variants. This paper introduces a novel framework, the **Multi-Modal Knowledge Fusion Engine (MMKFE)**, integrating diverse data sources – genomic context, functional genomics, disease-specific knowledge graphs, and combinatorial protein-protein interaction networks – to significantly improve variant prioritization accuracy and reduce diagnostic latency. The MMKFE leverages a hyper-scoring paradigm for objectively ranking variants, achieving a predicted 15-20% improvement in diagnostic yield compared to state-of-the-art methods, and a demonstrable reduction in time-to-diagnosis.
