FORMULATION AND EVALUATION OF TRANSDERMAL PATCHES OF LINAGLIPTIN HYDROCHLORIDE

A transdermal patches is medicated adhesive patches placed on skin to deliver a time release of medication through the skin for treating topical or systematic illness. Recent approach to drug delivery is to deliver the drug into systematic circulation at predetermined rate using skin as a site of application. Linagliptin is selected as a model drug for the present study. It exhibits the mechanism of action by inhibiting (DPP-4) enzyme for hypoglycemic activity. The oral bioavilabilty of linagliptin is approximately 30% thought linagliptin has a long terminal half life more than 100 hr. It exhibit accumulation half life approximately 10 hr to attend the steady state. The aim of present work was to prepare transdermal therapeutic system using various polymers, evaluate the physiochemical properties, nature of drug in formulation and in-vitro drug release through semi permeable membrane. The transdermal patches of Linagliptin were prepared by solvent casting method. The prepared formulation were prepared by varrying the concentration of polymer. The prepared formulation were evaluated for various parameter like thickness, weight variation, %moisture content, %moisture uptake, % drug content, in-vitro drug release and drug excipient compatability. Result: The formulation which (F3/F4) containing Linagliptin, ethylcellulose and PVP as a matrix forming agent and PEG 400 as plastisizer was the best formulation (F4). The in-vitro release data was treated with kinetic equation and it followed zero order release. The diffusion study was carried out by using egg membrane and showed 90.31% drug was release with in 10hr. Conclusion: The study conclude that transdermal patches can extend the release of linagliptin for many hour with the help of polymer ethylcellulose further it is also help in avoiding first pass metabolism.
