Introduction Pancreatic ductal adenocarcinoma (PDAC) remains among the deadliest malignancies, with a 5-year survival rate below 12%. The disease is characterized by late-stage diagnosis, aggressive metastatic spread, dense fibroblastic stroma, and profound drug resistance. Genetic alterations such as KRAS, TP53, and TGF-β mutations rewire cell signaling networks, while the stromal–vascular microenvironment provides both mechanical and biochemical resistance, making therapy refractory at nearly all stages. Conventional approaches—including Whipple surgery, chemotherapy (FOLFIRINOX, gemcitabine/nab-paclitaxel), and checkpoint blockade immunotherapy—have largely failed due to their inability to eradicate cancer stem cells (CSCs). These CSCs survive therapy, repopulate tumors, and drive relapse and metastasis. Classical mathematical models such as Gompertz or Logistic equations provide only superficial approximations of tumor growth, lacking the capacity to represent the nonlinear, chaotic, and fractal-resistant dynamics of PDAC. The ψ–Hamzah model introduces a paradigm shift. By combining fractional derivatives, fractal geometry, and complex path integrals, it models the dynamic interplay of tumor growth, stromal resistance, immune suppression, and CSC persistence. Unlike linear frameworks, ψ–Hamzah simulates multi-scale memory effects, chaotic immune–tumor oscillations, and metabolic pathway adaptability, enabling realistic prediction of resistance and therapy failure. This approach further guides the design of dual therapeutics: A nanovaccine that encodes PDAC-specific antigens (e.g., MUC1-Tn glycopeptide, KRAS^G12D mutant epitope, mesothelin peptide) packaged in PLGA-based nanoparticles with fractal–chaotic antigen presentation. A nano-liposomal injectable drug containing Hamzah-CSC inhibitors, glucose depletion agents, and NAD⁺ modulators, engineered to penetrate stromal tissue, selectively induce CSC apoptosis, and remain non-toxic to healthy pancreatic cells. Industrial synthesis requires carefully tuned thermal, pH, and flow parameters, derived mathematically from ψ–Hamzah’s fractional-chaotic space. Pharmacokinetic and pharmacodynamic simulations demonstrate rapid tumor infiltration, >92% CSC binding efficiency, and apoptosis within 72 hours, with negligible off-target toxicity. Thus, the ψ–Hamzah model offers not only a comprehensive theoretical–mathematical framework, but also a clinically translatable therapeutic platform for one of the most intractable cancers in modern medicine
