RAS proteins are small intracellular GTPases that regulate a variety of cellular processes, including proliferation, survival, differentiation, apoptosis, and cell-cell interactions. RAS proteins function by cycling between an active, GTP-bound state (RAS-GTP) and an inactive, GDP-bound state (RAS-GDP). In their active GTP-bound form, RAS proteins can bind and activate multiple effector proteins. Mutations in genes belonging to the RAS family are commonly found in 20%-30% of all human cancers, making them attractive targets for oncological therapy. During my thesis internship, I focused on studying the NRAS gene, a member of the RAS family, which is mapped to chromosome 1. Melanoma and multiple myeloma are among the tumors with the highest frequency of NRAS mutations. As an experimental model, the SK-Mel-2 melanoma cell line was used, which harbors the NRAS 182A>G (Q61R) mutation in heterozygosity. This specific mutation leads to the oncogenic activation of NRAS, resulting in cell proliferation that is independent of growth factors. Consequently, NRAS Q61 mutations are recognized as driver events in tumorigenesis and crucial therapeutic targets. However, the development of NRAS-selective inhibitors has not been successful in recent decades. For this reason, we investigated whether a gene-editing approach based on the CRISPR/Cas9 system could represent an alternative to conventional drugs. This approach relies on a gene-editing system composed of a guide RNA, complementary to the NRAS sequence, and a Cas9 protein that acts as a "molecular scissor" to cut the target gene. In this study, two mutated variants of Cas9, Cas9 EQR and Cas9 VQR, were utilized. These variants specifically recognize the PAM sequences "NGAG" and "NGA," which are exclusively generated in the mutant allele but not in the wild-type allele. As a positive control for NRAS gene silencing, previously selected siRNAs were used. These molecules efficiently inhibit the mutant NRAS variant by binding to its mutated mRNA and inducing degradation via the RISC complex. To assess successful editing of the mutant NRAS allele, molecular assays such as ASO-PCR, Sanger sequencing, and protein expression analysis via western blot were performed. Additionally, to determine whether NRAS inhibition had a biological effect, cell proliferation was evaluated through growth curve analysis and doubling time measurement.
