Recently, natural antioxidants are in growing demand for food, healthcare, and cosmetics industries. In addition, inedible mushrooms have been identified as potential sources of novel bioactive natural products. In this study, we found antioxidants present only in the fruiting body of the inedible mushroom Asterophora lycoperdoides, but not in its mycelia, using a TLC assay based on DPPH radical scavenging activity. Therefore, we attempted to isolate these antioxidants indicated by TLC colorization using column chromatography and HPLC. Two antioxidative compounds were isolated and identified as known compounds ergothioneine (1) and brunnein C (2) based on literature data from previous studies. Notably, in this study, the complete 13C NMR spectra of compound 2 were obtained for the first time. Compounds 1 and 2 demonstrated antioxidant activity in an in vitro DPPH radical scavenging activity assay, with compound 2 showing particularly potent activity. These results indicate that inedible mushrooms are underexplored sources of new natural antioxidants with various applications.
