Comparative Thalamic Effects of LSD, DMT, and Psilocybin: A Neuropharmacological Perspective
Inspired by recent work of Fang on the role of the thalamus, indicating that intralaminar and medial thalamic nuclei gate conscious perception, we present a comparative analysis of the effects of three classic serotonergic psychedelics lysergic acid diethylamide (LSD), N,N-dimethyltryptamine (DMT), and 4-phosphoryloxy- N,N-dimethyltryptamine (psilocybin) on thalamic function. Despite shared pri- mary receptor targets, these substances produce distinct phenomenological and neurophysiological profiles due to differences in onset kinetics, receptor binding affinity, and downstream modulation of thalamocortical connectivity. Special at- tention is given to alterations in sensory gating, signal entropy, and the dynamic reorganization of thalamic nuclei.
