Lactobacillus gasseri TF08-1 ameliorate high-fat diet induced Nonalcoholic fatty liver disease
The therapeutic options and efficacy for non-alcoholic fatty liver disease (NAFLD) are limited, and dysbiosis of specific gut microbiota is closely associated with the progression of this condition. In our previous study, we isolated Lactobacillus gasseri TF08-1 with probiotic potential from the feces of healthy adolescents. We successfully constructed the high-precision complete genome map of L. gasseri TF08-1 for the first time, and the annotation of this genome map showed an enrichment of lipid metabolism pathways. To investigate the therapeutic potential of L. gasseri TF08-1 in NAFLD, mice were given a high-fat diet (HFD) for 14 weeks and then supplemented with L. gasseri TF08-1 by gavage for 8 weeks. The results showed that in HFD-fed mice, the addition of L. gasseri TF08-1 significantly reduced lipid accumulation, improved hepatic steatosis, decreased levels of pro-inflammatory cytokines, and mitigated the progression of HFD-induced NAFLD. Metagenomic sequencing of fecal samples from the mice indicated that L. gasseri TF08-1 corrected HFD-induced gut microbiota dysbiosis, significantly increasing the abundance of beneficial bacteria such as Faecalibacterium prausnitzii and Phocaeicola vulgatus, while also enriching fatty acid degradation pathways. These findings suggest the probiotic potential of L. gasseri TF08-1 and provide a novel alternative for the treatment of NAFLD.
