High efficient chromatin conformation capture without pre-enrichment (HiChew) in single cells
Our method, HiChew (High efficient chromatin conformation capture without pre-enrichment), stands out for its high efficiency. What makes HiChew unique is that it doesn't require pre-enrichment before amplification. Instead, we use a high-efficiency sticky end ligation method (similar to Dip-C, HiRes), which minimizes the number of steps needed before amplification. After amplification, we label the ligation scar motifs with methyltransferase and enrich the ligation scars using a methylation antibody. Overall, our method has achieved the highest efficiency for single-cell Hi-C to date. This not only improves single-cell Hi-C, but also simplifies conventional Hi-C in common labs.
