Biological Pulse: A CRISPR-Cas13a-Based Salivary Biosensor Design for Subclinical Inflammation Detection
This project introduces a simulation-based CRISPR-Cas13a biosensor engineered to detect inflammatory biomarkers (IL-6 and TNF-α) in human saliva. The biosensor design uses a synthetic gene circuit hosted in a non-pathogenic bacterial chassis, modeled using tools like Benchling, RNAfold, and basic signal kinetics in Python. The study focuses on in silico prediction of biosensor behavior, covering: Biomarker specificity Signal activation time Environmental stability Safe oral microbial chassis use While the model has not been wet-lab validated, the data includes: 1.Simulated sensitivity (~95% for IL-6) 2.Genetic circuit diagrams 3.Stepwise biosensor workflow 4.Predictive tables (S1–S9). You are welcome to reuse, build upon, or test this system under open science principles. Supplementary material is included for reproducibility and future lab-based extensions.
